Question: I want to do a test run with my sample using the Chromium Nuclei Isolation Kit, do I need to use RNase Inhibitor?
Answer:
- For nuclei prep optimization purposes, the RNase Inhibitor is not required. PBS can be used as an alternative.
- However, if checking RNA quality post nuclei isolation or if wanting to do a pilot with single cell reagents, then an RNase inhibitor is required for Gene Expression experiments.
- Note that visual inspection of isolated nuclei will only provide readouts of nuclei yield, clumping, and debris. Visual inspection does not indicate data quality or single-cell behavior, a sequencing readout will provide this information.
