Question: What if my tissue is not on the list of tissues tested with the Chromium Nuclei Isolation Kit?
Answer: The Chromium Nuclei Isolation Kit has been tested on an extensive list of human and mouse tissues, testing both simple and challenging tissue types. The Chromium Nuclei Isolation kit has successfully isolated nuclei from many challenging tissue types, such as:
- Debris heavy tissues (e.g. Brain)
- Fibrous, challenging to dissociate tissues (e.g. Heart)
- RNase-rich tissues (e.g. Pancreas, Spleen)
The list of tissues tested reflect our access to these sample types and the specific tissues tested were chosen to reflect the wide range of possible input tissues expected to be used with this kit.
If your tissue of interest is not present in the list of tissues tested, other tissues which have similar characteristics may have already been tested and are compatible. For example, perhaps your tissue is fibrous and hard to dissociate, we have successfully isolated nuclei from mouse heart, which is also fibrous and hard to dissociate. If working with a high RNase tissue, we have successfully isolated nuclei and generated Gene Expression data from pancreas and spleen, which are both high RNase content tissues. This may be able to give you an indication of how a specific tissue may perform with the Nuclei Isolation kit and any general recommendations for those categories of tissues.
For untested tissues, users may wish to perform a pilot run before scaling up to a large number of samples.
