Question: How often should I refresh my deparaffinization and H&E staining solutions?
Answer: It may be possible to re-use solutions for xylene, graded ethanol, and H&E dyes when performing the Deparaffinization, H&E Staining, Imaging & Decrosslinking, or Deparaffinization, Decrosslinking, Immunofluorescence Staining & Imaging Demonstrated Protocols for Visium Spatial Gene Expression for FFPE.
If utilizing staining dishes, we recommend replacing solutions after one month or 20 slides (whichever comes first). If utilizing 50 ml conicals or Coplin jars, we recommend replacing solutions after two weeks or ten slides (whichever comes first).
To effectively re-use solutions, be sure to do the following:
- Rotate them periodically to keep them fresh (e.g., swap Xylene Jar 1 and Xylene Jar 2).
- After use, transfer the solutions to a tightly sealed container to prevent air exposure.
