Question: How do I assemble HT Gel Beads in the HT Gel Bead holder?
Answer: Ensure that the gel bead strip is positioned with one tube in the left-most position (do not center the strip if using fewer than 8 tubes). Gently depress the lid until light resistance is met. Do NOT attempt to depress the lid further, even if this results in the lid sitting askew on the strip holder.
When opening HT Gel Bead packaging, you will see the following for either the 48 reaction kit (left) or the 8 reaction kit (right):
After gel beads are thawed for at least 30 minutes at room temperature before use, snap the tube strip holder with the Gel Bead strip into a 10x Vortex Adapter. Vortex for 30 seconds.
Centrifuge the Gel Bead strip for ~5 sec after removing from the holder. Confirm there are no bubbles at the bottom of tubes and the liquid levels look even. Place Gel Bead strip back in the holder and secure the holder lid.
For both full stips of 8 gel bead aliquots (48 reaction kit, left) and partial strips of 4 gel bead aliquots (8 reaction kit, center), only depress the lid until light resistance is met. There should be a gap between the black lid and the clear plastic gel bead holder. Do not depress the lid too far, so there is no gap visible (right). For partial strips, such as is the case for the 8 reaction kit, make sure to center the Gel Bead strip to the left, as shown above.
Forcing the lid for HT risks the following:
- The lid is very difficult to remove
- Gel bead tubes are crushed
- Foil is punctured
