Question: How does the data compare between fresh and frozen samples in the Multiome assay?
Answer: We compared the data from fresh and cryopreserved-thawed human PBMCs from the same individual. We noticed a good separation between cell and non-cells and good ATAC targeting metrics. The fraction of transcriptomic reads in cells and the number of genes detected were comparable between the fresh and frozen-thawed PBMCs. The fraction of different cell types was also conserved between fresh and frozen samples.
We have also tested E18 mouse brain from the same mouse as a fresh sample and a flash-frozen sample using the Multiome assay. We found a good separation of cells and non-cells in the cross-sensitivity plot and a comparable number of genes were detected. We saw a slight reduction in ATAC targeting metrics in the frozen sample. Similar cell populations could be identified in both fresh and frozen samples.
