Question: Which genes does each V(D)J-specific primer map to?
Answer: The V(D)J primer sequences used for enrichment are specifically targeted towards the constant region of the T- or B-cell transcripts. This region is constant across multiple transcripts, which allows us to capture multiple variants with just a few specific primers. We use a nested-PCR enrichment method in order to increase specificity of the amplification product.
