Question: If I am running a time-course study collecting tissues, what is the best way to section and prepare the Visium Gene Expression slide with tissue?
Answer: We would recommend placing all four tissue sections on a Visium slide in a single cryosectioning event. Prepare tissue blocks for each time point by snap freezing with an isopentane and liquid nitrogen bath. Place tissue block in pre-chilled sealed container and store at -80C until all time point blocks have been collected and properly snap frozen. Then embed the individual blocks in OCT. Perform sectioning when all the blocks are snap frozen and embedded in OCT and prepare the slide with individual tissue sections. We recommend changing blades between each tissue type. The Visium slide with tissue sections placed can be stored in a sealed container at -80C for up to 4 weeks.
