Question: Is it possible to freeze nuclei prior to Chromium Single Cell assays?
Answer: We do not recommend freezing isolated nuclei prior to performing Chromium Single Cell assays. The freeze/thaw process can compromise the nuclear membrane, leading to a loss of single-nuclei behavior. This approach is untested and unsupported. Therefore, we cannot provide specific guidance.
Instead of freezing isolated nuclei, we recommend alternative strategies. For more information, please see our Cell Preparation for Single Cell Protocols Handbook.
If isolated nuclei must be stored, nuclei suspensions can be fixed following the Fixation of Cells & Nuclei Demonstrated Protocol for downstream Flex Gene Expression assays.
