Question: What is the difference between Amp Mix (PN-2000047) and Library Amp Mix (PN-2000531)?
Answer: Amp Mix and Library Amp Mix have different formulations. Because of the formulation differences, Amp Mix is suitable for all (cDNA and library) amplification steps in the GEM-X 3’ v4 or 5’ v3 workflows while Library Amp Mix is suitable for only specific library amplification steps. Amp Mix and Library Amp Mix are provided in different kits and should be used as per GEM-X User Guide instructions. See below for details.
Library Amp Mix is only included in the Library Construction Kit C, 16 rxn (PN-1000694). Library Construction Kit C, 4 rxn (PN-1000689) does not include Amp Mix or Library Amp Mix as sufficient Amp Mix is included in the GEM Kit to complete the workflow.
Amp Mix may be used for the following steps:
- cDNA Amplification
- V(D)J Amplifications (for 5’ v3)
- Gene Expression Sample Index PCR
- V(D)J Sample Index PCR (for 5’ v3)
- Feature PCR (for 5’ v3 CRISPR)
- Feature Barcode Sample Index PCR
Library Amp Mix should only be used for the following steps:
- Gene Expression Sample Index PCR
- V(D)J Sample Index PCR (for 5’ v3 only)

Incorrectly using Library Amp Mix when the User Guide calls for Amp Mix will lead to an impact on assay performance.
