Question: Can I use a Cassette Lid instead of PCR film during Cell Segmentation Staining?
Answer:
The answer depends on the assay chemistry.
Xenium In Situ Gene Expression v1
No. The current Xenium v1 Cassette Lid does not fit well with the Xenium Cassette insert. Using a Xenium v1 Cassette Lid instead of PCR film during the staining workflow risks increased condensation during overnight staining as well as decreased assay performance. For optimal assay performance, use the provided PCR film.
Xenium Prime In Situ Gene Expression
Cell Segmentation Staining should be performed with a Xenium v2 Cassette Lid as described in the User Guide. Using PCR film during the Xenium Prime assay workflow risk evaporation and decreased assay performance. Discard the PCR film provided with the Cell Segmentation Staining Reagents.
Whether you are performing overnight staining with Xenium v1 (PCR film) or Xenium Prime (cassette lid) reagents and consumables, staining is performed at 4C in a fridge or incubator.
