Question: How do I study large tissue sections using Xenium?
Answer: The imageable area on a Xenium slide is 12mm x 24 mm with an optimal area for sample placement, so as not to cover fiducial markers, of 10.45mm x 22.45mm. For details on the fiducial frame and sample placement area, see: How can I more easily visualize the fiducial frame on Xenium slides?
Depending upon the tissue type, scoring of the tissue may be employed if the tissue is too large to fit within the imageable area. Multiple scored regions of the same block can be placed on a Xenium slide capture area as long as the sections (and corresponding embedding media - paraffin or OCT) do not overlap. In data analysis, each region produces a separate output bundle. If you were to separate tissues that actually need to be combined, the dataset would somehow need to be rejoined. This is officially unsupported by 10x Genomics.
