Question: Can I use 100% Tween 20 diluted to 10% Tween 20 for Xenium?
Answer: Tween 20 Surfact-Amps Detergent Solution (10% solution) (Thermo Fisher Scientific, PN 28320) is used in the Xenium assay and instrument workflow. It is strongly recommended to use the Tween 20 part number listed in the Demonstrated Protocols and User Guides.
Substituting 100% Tween 20 (diluted to 10%) may adversely affect assay and system performance and therefore is not recommended. The viscosity of 100% Tween leads to measurement inaccuracies and variable concentration in the final solution. If the validated Tween 20 cannot be obtained, it may be possible to substitute additional reagents as low-risk alternatives as long as they meet the following criteria:
- 10% Tween v/v
- Same molecular biology grade and chemical formulation
- Nuclease-free
If using 100% Tween 20 (not recommended and unsupported), only measure using positive displacement pipette to dilute.
