Question: How do I determine if my tissue block will be compatible with Xenium?
Answer: We recommend morphological assessment of tissue blocks of interest by DAPI and H&E QC before Xenium In Situ Gene Expression. DAPI and H&E QC guidelines can be found in the Tissue Preparation Guides for both Fresh Frozen and FFPE tissue samples.
We have found that tissue morphology rather than DV200 or RIN is better correlated with decoding metrics. However, we recommend tissues used in Xenium In Situ assays should have a DV200 of >30% as discussed in Is RNA quality assessment (either by RIN or DV200) important for Xenium?
