Question: Is there a detection bias for very short or long transcripts?
Answer: The Single Cell 3' assay can detect polyadenylated RNA up to at least 20 kb in length.
Very short transcripts (<200 bp) will be removed during the SPRI cleanup. However, those are typically the result of degraded RNA molecules.
Any amplification bias introduced during cDNA amplification or the Sample Index PCR will be corrected with the Unique Molecular Identifiers (UMIs) during data analysis.
