In short, we recommend that organoid tissue should be dissociated into single cell suspensions. Preparation of single cells from organoids could be achieved by using different enzymes such as Accutase®, Papain, Trypsin, TrypLE™ or Dispase. Optimization of enzyme digestion time should be considered in order to obtain healthy single cells during dissociation steps.
The single cell suspensions can then be fixed following the Fixation of Cells & Nuclei Demonstrated Protocol for use with Flex Gene Expression assay.
