Question: What are best practices for working with organoids in the FRP?
Answer: Our recommendations for working with organoid tissue for 10x single-cell assays can be found here: Are there any recommendations for working with organoid tissue for 10x single cell assays?
In short, we recommend that that organoid tissue should be dissociated into single cell suspensions. Preparation of single cells from organoids could be achieved by using different enzymes such as Accutase®, Papain, Trypsin, TrypLE™ or Dispase. Optimization of enzyme digestion time should be considered in order to obtain healthy single cells during dissociation steps.
The single cell suspensions can then be fixed following the Fixation of Cells & Nuclei for Chromium Fixed RNA Profiling Demonstrated Protocol for use with Fixed RNA Profiling assay.
