While the fill volume on the label reads 250µL, the actual fill volume is 270µL to help compensate for variability in user pipetting. The total fill volume is 1350µL ( 5 tubes x 270µL). For 16 reactions, customers will use a total of 1320µL. This volume already includes the 10% overage to make master mixes comfortably, meaning you still have 30µL left over for pipetting errors on top of the extra 10%.
