Question: How is tissue preparation similar and different between Visium for FFPE (v1) and Visium CytAssist for FFPE?
Answer: Both chemistry versions utilize human or mouse FFPE samples as inputs. For the CytAssist assay, FFPE blocks or archived sections on Superfrost glass slides can be used.
For the Visium for FFPE product, section placement and on-slide workflow steps occur on the Visium Spatial Gene Expression Slide. In contrast, the tissue used for the CytAssist for FFPE assay is placed on plain glass slides, and probes are transferred to the Visium CytAssist Spatial Gene Expression Slide by the Visium CytAssist instrument.
For the Visium for FFPE assay, larger tissues or blocks should be trimmed or scored to fit within the capture area. For the CytAssist for FFPE assay, if using larger tissues or blocks, they do not need to be trimmed.
The area of interest will need to be within the compatible region of the slide, centered within the gasket of the Tissue Slide Cassette, and centered within the alignment guides on the Tissue Slide stage.
