Question: Which thermal cyclers are compatible with the Visium for FFPE v1 assay?
Answer: The following thermal cyclers have been validated with the Visium for FFPE v1 assay. Validated thermal cyclers have undergone rigorous testing and have generated reproducible, high-quality results when following our 10x Genomics Visium for FFPE v1 Tissue Preparation Guide, Staining Demonstrated Protocols, and User Guide. Please note: we are currently in the process of updating our documentation to include the Eppendorf Mastercycler X50s and Biometra TAdvanced thermal cyclers, which have been recently validated with the Visium for FFPE assay.
Thermal cyclers that are not included in the list above are unsupported as we cannot guarantee assay performance. Untested thermal cyclers may be incompatible with the Thermocycler Adaptor and/or Slide Cassette, which can lead to cassette leakage, broken slides, and the possibility of generating suboptimal data as compared to validated thermal cyclers. For instance, we have found that the Visium Thermocycler Adaptor doesn’t sit flush with the Bio-Rad T100 thermal cycler block. Based upon limited testing with Visium for Fresh Frozen assay, we observed a decrease in sensitivity (i.e., Genes and UMIs per Spot) and mapping rates when comparing the data generated using a Bio-Rad T100 to a validated thermal cycler. The observations made with the Visium for Fresh Frozen assay likely translate to non-optimal performance for the Visium for FFPE v1 assay as well.
We would recommend that Visium customers use a validated, supported thermal cycler for performing their Visium experiments for optimal assay performance.
