Question: What are the best practices for tissue dissociation using the Chromium Nuclei Isolation Kit?
Answer: Dissociate the tissue in the Lysis Buffer by grasping the pestle between thumb and fingers. Twist the pestle continuously within the tube. Alternate between pushing the pestle down and lifting the pestle up to allow the tissue to slide between the wall of the tube and the pestle. Continue dissociating the tissue until the lysate appears homogeneous and no large pieces of tissue remain.
