Question: Will the nuclei isolated using the Chromium Nuclei Isolation Kit be compatible with Feature Barcoding, CellPlex, CRISPR screening, or VDJ profiling?
Answer: The process of nuclei isolation removes outer cell membranes and cytoplasmic compartments, therefore:
- Feature barcoding for cell surface protein or antigen specificity is not compatible with nuclei.
- VDJ capture from nucleic transcripts is challenging and not currently supported.
The use of nuclei from frozen tissue with CellPlex is not currently supported. Nuclei isolated from fresh tissue with the Chromium Nuclei Isolation Kit are untested and unsupported with CellPlex.
Cultured cells are not compatible with the Chromium Nuclei Isolation Kit. Cultured cells are required for the transduction of CRISPR libraries and subsequent antibiotic selection, and therefore the kit is not currently compatible with CRISPR screening.
