Question: What cell filters or strainers are recommended for use with Fixed RNA Profiling assay?
Answer: Cell filters are recommended at multiple steps within the Fixed RNA Profiling workflow.
Following sample fixation, fixed single cell or nuclei suspensions can be filtered using a 30 μm filter (Sysmex CellTrics or Miltenyi Biotec Pre-Separation) to remove cell aggregates or clumps.
Following the post-hybridization washes, we strongly recommend that pooled samples be filtered with a 30 μm filter (Sysmex CellTrics or Miltenyi Biotec Pre-Separation) before loading the sample onto the microfluidic chip (i.e., Chip Q).
Best Practices using the 30 μm filters:
- Hold the pipet tip at an angle and touch the filter membrane with the tip where the filter meets the wall. Slowly pipet through the filter.
- Tap gently or spin down if liquid remains at the end of the filter.
Residual volume can be pipetted from underneath the filter to maximize cell recovery.
If these validated filters are not available, alternative filters can be used as long as they are 30 μm AND rated for the concentration range of cells (500-8000 cells/uL).
