Question: How many Feature Barcode kits do I need for the HT assay?
Answer: Each HT reaction requires two Feature Barcode reactions to complete. This is because the GEMs are split into two tubes as part of the Feature Barcode library preparation workflow. Therefore, cDNA and Feature Barcode DNA amplification has twice the number of tubes as the standard assay and requires twice the volume of primers. See below for sample calculations for Feature Barcode kit purchase:
3' HT v3.1:
- One 3’ Feature Barcode 16 rxn kit (PN-1000262) is required for processing 8 samples.
- Two 3' Feature Barcode 16 rxn kits (PN-1000262) are required for processing 16 samples.
- Six 3' Feature Barcode 16 rxn kits (PN-1000262) are required for processing 48 samples.
5' HT v2:
- One 5’ Feature Barcode 16 rxn kit (PN-1000541) is required for processing 8 samples.
- Two 5' Feature Barcode 16 rxn kits (PN-1000541) are required for processing 16 samples.
- Six 5' Feature Barcode 16 rxn kits (PN-1000541) are required for processing 48 samples.
