Question: What sample preparation recommendations should be considered when using the HT solution?
Answer: The Next GEM Single Cell HT assays are compatible with the same sample and tissue types as current on-market 3’ and 5’ applications.
- Because of the high cell loads, we recommend following the Sample Preparation recommendations outlined in our Best Practices to Minimize Chromium Next GEM Chip Clogs and Wetting Failures Technical Note and Cell Preparation Guide. Using the same sample preparation techniques will ensure similar assay performance between our standard assay at high cell loads and our HT assay. Variations in sample preparation, such as excess debris, dead cells, or large cells can result in performance differences.
- It is important to note that clogs and wetting failures are highly dependent on sample quality. If suboptimal samples are used (e.g., suspensions that contain debris, cell aggregates, dead cells, or cells larger than 50 microns), the risk of obtaining a clog or wetting failure may increase as more volume of the single-cell suspension is loaded onto the chip. Variations in sample preparation can also result in performance differences.
