Question: If there is a mutation within the binding regions of two probes, will the probes still bind to the mRNA?

Answer: Variation in the target is tolerated but some positions are better tolerated than others. We have observed that ligation efficiency does go down if there is a mismatch at the ligation junction in the probe.
