Question: What tissue section thickness is recommended with the Visium HD 3' Spatial Gene Expression assay?
Answer: For optimal assay performance with Fresh Frozen, OCT embedded tissues, we recommend a section thickness of 10 μm.
- Obtaining high-quality tissue sections thinner than the recommendation can be challenging due to difficulty during sectioning, resulting in a higher frequency of folds and artifacts. These artifacts and folds increase risk of tissue detachment and further suboptimal assay performance.

Processing sections thicker than the recommendation will likely result in lower assay performance. We have observed a significant reduction to sensitivity (UMI Counts) & decrease in Mapping (Reads Mapped Confidently to Transcriptome) when processing ≥15 μm sections.
