Question: How do I exclude RBCs from my cell count?
Answer: If no RBC lysis is performed, red blood cells will make up part of the total sample. This is an important factor to consider when trying to achieve accurate cell recovery. Any remaining RBCs may be included in the final cell count and calculations for sample loading.
A fluorescent dye such as AO/PI can be implemented to exclude RBCs from the final cell count. RBCs, platelets, and debris will not fluoresce and not be included in the final cell count.
