Question: How do I know my RBC lysis is working?
Answer: The turbidity of the sample can assess red blood cell lysis efficiency. If starting with whole blood, the sample will go from opaque (no light goes through) to clear. If the sample is still very turbid after the first RBC incubation, the RBC lysis can be repeated.
The images below show the progression of a whole blood sample after two rounds of RBC lysis.
