Question: Can I use 1.5 ml tubes instead of 0.2 ml tubes for vacuum centrifugation in the Targeted Gene Expression assay?
Answer: We do not recommend drying down libraries in 1.5 ml tubes in the vacuum centrifuge. We recommend using a vacuum centrifuge that accommodates 0.2 ml tubes.
If a vacuum centrifuge that accommodates 0.2 ml tubes is not available, we recommend using SPRIselect reagent to concentrate libraries, as outlined in our Demonstrated Protocol.
If you choose to use a vacuum centrifuge that accommodates 1.5 ml tubes, it will be necessary to transfer the samples (after resuspending the dried libraries in Hybridization Master Mix) to 0.2 ml tubes for the subsequent hybridization step. It is likely that some library material will be lost during this transfer step, due to adsorption to the 1.5 ml tube. It may be beneficial to add an extra PCR cycle to the post-capture library amplification step, to account for the loss in material.
