Question: Can I use Dual Index Plates together with 3' v3.1 Single Index reagent kits?
Answer: If you have already purchased 3' v3.1 Single Index reagent kits, it is possible to upgrade to Dual Indexing by following these steps:
- For generating Dual Index Gene Expression libraries:
- Purchase the Dual Index Kit TT Set A
- Use this Dual Index plate together with your existing 3'v3.1 chips, gel beads, and reagents, following the User Guide for 3' v3.1 Dual Index Gene Expression libraries.
- Use Dual Index Plate TT (instead of the Single Index Plate T and the SI Primer)
- Please ensure that you do not use the SI Primer when generating Dual Index libraries.
- For generating Dual Index Gene Expression and Cell Surface Protein libraries:
- Purchase the Dual Index Kit TT Set A, and the Dual Index Kit NT Set A
- Use these Dual Index plates together with your existing 3'v3.1 chips, gel beads, and reagents, following the User Guide for 3' v3.1 Dual Index Gene Expression and Cell Surface Protein libraries.
- For Gene Expression libraries, use Dual Index Plate TT (instead of the Single Index Plate T and the SI Primer)
- For Cell Surface Protein libraries, use Dual Index Plate NT (instead of the Single Index Plate T and the Feature SI Primers 2)
- For generating Dual Index Gene Expression and CRISPR Screening libraries:
- Purchase the Dual Index Kit TT Set A, the Dual Index Kit NT Set A, and the 3' Feature Barcode Kit
- Use these Dual Index plates and Feature Barcode kit together with your existing 3'v3.1 chips, gel beads, and reagents, following the User Guide for 3' v3.1 Dual Index Gene Expression and CRISPR Screening libraries.
- For Gene Expression libraries, use Dual Index Plate TT (instead of the Single Index Plate T and the SI Primer)
- For CRISPR Screening libraries, use Feature SI Primers 3 (instead of Feature SI Primers 1) and the Dual Index Plate NT (instead of the Single Index Plate T and the Feature SI Primers 2). Please note that the PCR conditions and SPRI ratios are different for single index and dual index CRISPR libraries.
