Question: I installed a custom sequencing recipe on my NextSeq 500/550, why can't I input 24 bp in the i5 indexing read?
Answer: The NextSeq 500/550 instrument software does not support an i5 indexing read of 24bp. In order to accurately capture and read the 10x barcode on Multiome ATAC libraries, the use of a custom sequencing recipe is required. After installing the custom recipe, the user must restart their instrument and select the custom recipe from the drop-down list during the sequencing run set-up. The user must input 16bp in the i5 indexing read, as the 8 dark cycles have already been accounted for in the custom recipe.
