Question: Can I purchase custom panels from other vendors for use in the Targeted Gene Expression assay?
Answer: While we recommend purchasing custom panels from IDT, or generating custom panels by following our Demonstrated Protocol CG000412 for biotinylation and amplification of Twist Oligo Pools, it may be possible to use custom panels purchased from other vendors. However, these are not officially supported by 10x Genomics as they have not been tested nor validated in our target enrichment workflow. Assay performance cannot be guaranteed.
General guidelines for working with alternative oligo providers:
- Custom panels should be purchased as single-stranded DNA oligonucleotides that have been individually synthesized, biotinylated at the 5' end, and pooled.
- Sequences for each individual oligonucleotide can be found in the Bait Design file that is output from the 10x Genomics Custom Panel Designer, and are ~120 nucleotides in length.
Adding genes to 10x pre-designed panels:
- Add-on panels should be diluted to a working dilution of 20 amol/probe/µl in 1x TE buffer, pH 8.0, and combined with a 10x pre-designed panel as described in Step 2.1B (p 29) in the Targeted Gene Expression User Guide.
Working with fully custom panels:
- Fully custom panels are diluted so as to ensure a sufficient amount of biotinylated bait in every pulldown reaction. Baits should be diluted with 1x TE buffer, pH 8.0 according to the table below. When working with products other than the Compatible Products from IDT listed in the User Guide, the table below should replace Table 1 (p 30) in the Targeted Gene Expression User Guide.
