Question: What tissue types require further optimization for use with Visium?
Answer: A subset of tissues we’ve tested in-house require additional optimization outside of the recommended tissue preparation methods and Visium workflows. These tissues include:
· human and mouse skin
· mouse femur
· mouse pancreas
Alternative sample preparation methods such as decalcification and cryosectioning blade type for bone tissue, section thickness and sectioning temperatures for fatty tissues, and/or longer permeabilization times may need to be explored. We are currently working on optimizing Visium for these tissue types, but unfortunately we do not currently have a timeline on when this information will be available. Other tissues we have not tested may also require more extensive optimization. Customers interested in working with untested or challenging tissue types should utilize the Tissue Optimization slide to optimize their conditions before proceeding with the Visium Spatial Gene Expression experiment.
