Question: Are there any safe stopping points in the Chromium Connect workflow?
Answer: There are no safe stopping points in the Chromium Connect workflow.
However, there are pauses for quality checks where the instrument, reagents, and samples are held at safe temperatures on the deck.
Pauses will occur at the following QC points in the workflow when these options are selected:
- GEM QC check (optional) - the instrument will pause for 5 minutes to allow for visual inspection of the GEMs. The user will then be prompted to enter the GEM QC data by selecting Pass/Fail from the drop-down menu for each sample.
- midrun QC (optional) - when this option is selected, the assay will pause after cDNA amplification to allow for the user to perform a cDNA QC. The cDNA used for QC is separate from the cDNA used for the subsequent library preparation steps. The user will be prompted to enter the cDNA concentrations for each sample.
- library QC - the final libraries are held at a safe temperature until the user ends the run.
