Question: Can the H&E staining process have a negative impact on RNA integrity and reduce assay sensitivity?
Answer: It is possible to have a certain decrease in RIN value at individual steps of the Visium workflow, but we have optimized the critical workflow steps (sectioning, fixation, storage and staining) not only to maintain the RNA integrity but also tissue morphology for optimal assay and application performances.
In order to ensure the best possible results, we recommend using tissue with a RIN (RNA Integrity Number) score of 7 or greater. To evaluate tissue we recommend performing an RNA quality assessment on a representative tissue block or the block you will be processing with Visium. Please refer to the Tissue Preparation Guide for more details.
