Question: When do I need to sequence my V(D)J-enriched libraries more deeply?
Answer: In most cases, our standard recommendations (5,000 read pairs per targeted cell in a 26 x 91 read length configuration) is sufficient. However, some sample types do not reach saturation at our recommendation sequencing depth and there are some instances that may require deeper sequencing. This may include the following scenarios:
- The sample contains many plasma B cells, which have roughly two orders of magnitude higher expression of IG genes than other B cells.

If recovery of low expression cells is critical to your experiment design, and your sample type is prone to high variation, use higher depth.
