Question: What should I do if I accidentally use Chip A instead of Chip B with Single Cell 3' v3 reagents?
Answer: As for all 10x Genomics solutions, the recommended chip kit is designed to be used with the corresponding reagent kit.
For Single Cell Gene Expression, the Single Cell Chip B has been optimized for performance with the Single Cell 3' v3 Reagent Kit. Single Cell Chip B has also been updated to include additional run features that aim to improve assay performance. Therefore, Single Cell 3' v3 Gene Expression (or Gene Expression and Feature Barcoding technology) should be run on Single Cell Chip B.
If Single Cell Chip A (which was developed for Single Cell 3' v2 and Single Cell V(D)J Reagent Kits) is accidentally used with Single Cell 3' v3 reagents, it will still be possible to generate GEMs and produce sequencer-ready final libraries.
However, note that the 'Estimated Number of Cells' detected by Cell Ranger may show a larger deviation relative to the number of cells that were targeted in the experiment. The data generated from each cell, however, is usable.
