Question: Do I need to dissociate tissues into single cells first before isolating nuclei, or can I use the whole tissue?
Answer: Tissues must first be dissociated into single cell suspensions. However, certain tissues (i.e. brain) do not readily dissociate. For those instances, it is possible to go straight into nuclei isolation. Guidance for nuclei isolation from tissues will be provided in a future demonstrated protocol.
