Question: Do I need to remove RNA from my genomic DNA sample?
Answer: The effect of residual RNA in genomic DNA samples when performing the genome or exome assay has not been examined. However, it is not expected to have a large effect because the first step in the assay is denaturation with NaOH, which should degrade any residual RNA.
The Demonstrated Protocols for isolating high molecular weight DNA for the Genome assay include workflows that both include RNase treatment (Qiagen Magattract) and do not include RNase treatment (salting out method) and both produce successful libraries.
