Question: Can frozen tissue be used for the Single Cell CNV workflow?
Answer: Yes, frozen tissue be used as input for Single Cell CNV workflow.
We recommend isolating nuclei directly from the frozen tissue. We have a protocol for isolating nuclei from snap-frozen tissue available here: https://support.10xgenomics.com/single-cell-dna/sample-prep/doc/demonstrated-protocol-isolation-of-nuclei-for-single-cell-dna-sequencing
Other tissues may require additional optimization such as increasing the number of pestle strokes (if the tissue is tough) or increasing the lysis time (if unlysed cells are observed).
Note: High starting sample quality is important. Specifically; how the tissue was dissected, frozen, and how long it sat out beforehand, will impact data quality.
