Question: My cell type is difficult to lyse. How can I test whether my cells will lyse in GEMs?
Answer: The Single Cell 3' and 5' workflows have been optimized for mammalian cells. Other cell types, such as plant cells, which contain a cell wall, may not efficiently lyse in the 10x assay. In these cases, one option is to isolate protoplasts. If you are concerned whether your cell type will lyse in our specific lysis buffer, we suggest attempting to lyse your cells in bulk with 0.8%, 1% and 1.2% Triton-X. If these concentrations of Triton-X are able to lyse your cells, then the cells will likely lyse in our workflow.
Note: It is not recommended to add Triton-X or any other detergent to the Master Mix to increase lysis yield as this may interfere with the GEM generation.
