Question: Can I store my amplified cDNA beyond 1 week at -20C?
Answer: We have not tested storing the amplified cDNA beyond 1 week at -20C. It is likely safe to store the amplified cDNA for up to two weeks, but this is something we have not confirmed. The risk of storing it longer is that there will be a lower number of genes detected per cell if the cDNA is degraded. The safest thing to do is to process the amplified cDNA within 1 week of storage at -20°C.
If possible, proceed to generating the final libraries as these can be frozen at -20°C for long term storage. In-house we've sequenced libraries that were stored for more than a year. Just make sure the libraries are re-quantified with the KAPA kit as some small degradation of the library may occur.
