Question: Are cardiomyocytes compatible with Single-Cell assays?
Answer: We have not tried processing cardiomyocytes internally but have had some customers attempt to process them with various success rates.
The maximum cell size we have tested in-house is 30 um. Larger cells such as cardiomyocytes may clog the microfluidic channels in the 10x Genomics chips.
One important distinction is whether the cardiomyocytes are from adult or embryonic samples and the organism. Some customers have successfully generated GEMs and amplified cDNA from mouse embryonic heart tissue. However, with older mice and adult patient cardiomyocytes, the larger size of the cells may clog the microfluidic channels on the chip. For adult cardiomyocytes, nuclear isolation may be a more reliable way to run these samples.
