Question: Can automated libraries be made on the Chromium Connect from GEM-X cDNA?

Answer: It is possible to use cDNA generated from GEM-X kits as inputs for automated library construction using the Chromium Connect but this would be an unsupported workflow.

GEM-X 3’ v4 and 5’ v3 Gene Expression Library Construction:

• Pipette 15 μl of purified cDNA (manually processed from Chromium X/iX, see GEM-X User Guides: CG000731 or CG000733) into the ODTC full-skirted plate column 7.  
• Acquire the following Automated Kit materials (LIT000154):
  • Automated Library Construction Kit
    • 4 rxns: PN-1000429
    • 24 rxns: PN-1000428
  • Dual Index Kit TT Set A
    • 96 rxns: PN-1000215
• Follow the instructions for Automated Gene Expression Library Construction outlined in CG000474 
  • For Sample Index PCR, refer to the relevant GEM-X User Guide (CG000731 or CG000733) for recommended cycle numbers
• Select Library Construction > Single Cell GEX Kit on the Chromium Connect screen
