Question: Are GEM-X Chips more robust to emulsion failure than Next GEM Chips?
Answer: GEM-X Chips have improved robustness to emulsion failures based on side-by-side testing against Next GEM Chips. This is enabled by new chip design, faster flow rates, and new reagents.
One of the primary drivers of sample clogs is the presence of cell aggregates and debris that negatively impact the fluidics in the 10x Chip. When running experiments, we strongly recommend utilizing sample handling best practices including:
- Implement straining/filtering steps
- Utilize a cell suspension that has high cell viability and minimal debris/aggregates under visual inspection
- Minimize time between generating a suspension and chip loading
