Question: Can dilute gDNA (<1 ng/ul) be used for Genome libraries?
Answer: If the concentration of HMW gDNA is less than 1 ng/ul but equal to or greater than 0.5 ng/ul, it is possible to proceed with the Genome workflow with modification. The gDNA sample should be denatured as written in the User Guide, but the volume of DNA/Denaturing Agent added to the Sample Master Mix should be increased, up to 5 ul total, for a total DNA mass of 1.25 ng. Below 0.6 ng, the performance of the assay may decline.
We haven't tested concentrating HMW gDNA samples with a DNA vacuum concentrator (e.g. SpeedVac) or centrifugation and pelleting. However, these methods may damage HMW gDNA due to heat and/or extra pipetting steps.
