Question: What is surrogate fluid?

Answer: Surrogate fluid is glycerol in a ~50% v/v aqueous solution. The 50% glycerol solution can be purchased from Ricca Chemical Company, Glycerin (glycerol), 50% (v/v) Aqueous Solution (PN-3290-32).

Alternatively, 50% glycerol solution can be made from a stock solution of glycerol as follows:

1. Mix an equal volume of water and 99% Glycerol, Molecular Biology Grade.
2. Filter through a 0.2 μm filter.
3. Store at −20°C in 1 ml LoBind tubes.
4. 50% glycerol solution should be equilibrated to room temperature before use.
