Question: How do I prepare and store 10% IGEPAL CA-630 and IGEPAL CO-360 stock solutions for nuclei isolations?
Answer: Many of our Demonstrated Protocols for nuclei isolations prepare the lysis buffer from 10% (v/v) IGEPAL stock solutions. For example, 100ul of 100% detergent would be diluted into 900ul nuclease-free H2O. Refer to the specific Demonstrated Protocol to determine whether IGEPAL CA-630 or IGEPAL CO-630 is recommended as these are different compounds.
After preparing 10% IGEPAL solutions, heating the solution at 40C for 10 minutes can increase the rate at which the detergent goes into solution. It may also be helpful to prepare 10% IGEPAL on the day before a planned experiment to let any bubbles settle out before use.
We have not conducted official stability tests. However, storing 10% IGEPAL solutions for several weeks should be low risk.
