Question: What can I use in place of Probe Dilution Buffer?
Answer: Probe Dilution Buffer (PN-2000393) is currently being utilized in the Xenium In Situ Gene Expression Assay for the following:
- Add-on or standalone custom probe resuspension (probe part numbers can be found here.
- Probe Hybridization Mix set-up (Step 1: Probe Hybridization, CG000582)
A single tube of approximately 750 ul is supplied with either base panel oligos or custom panel oligos.
Probe Dilution Buffer has been removed from the Assay User guide and will be removed from Xenium Panel Kits at a later date.
Instead, customer-supplied TE Buffer, TRIS-EDTA, 1X Solution, pH 8.0 Thermo Fisher BP24731 (nuclease-free)(Thermo Fisher Scientific BP24731) will be used in place of Probe Dilution Buffer. This change applies to both custom probe resuspension and probe hybridization mix set-up. What alternative TE buffers can be used for Xenium?

Kitting changes take time and therefore in the short-term, panel kits will be supplied with a tube of Probe Dilution buffer. This tube can be discarded or can be used in place of TE buffer if sufficient volume is available. Please note that a single tube of Probe Dilution Buffer is approximately 750 ul. Approximately 371 ul of TE is required for Probe Hybridization and 625 ul is required for add-on custom or standalone custom oligo resuspension.
