Question: How do I determine if my tissue block or archived section will be compatible with the Visium CytAssist Spatial Gene and Protein Expression assay?
Answer: To assess block performance with the Gene Expression component, we are recommending that customers perform an RNA Quality Assessment of their tissue block of interest or scraping a representative section from an archived slide before the Visium Spatial assay. Based on in-house experiments, we recommend using tissue with DV200% ≥30%. Using low DV200-scored blocks will still yield data but likely of lower sensitivity (i.e., lower UMIs and Genes per Spot). Details on how to perform RNA Quality Assessment are in our Tissue Preparation Guide.
To assess likely block performance with the Protein Expression component, you can optionally perform an immunofluorescence staining using antibodies found in the Human Immune Cell Profiling Panel. Antibody clones in the 10x Genomics Antibody Panel are listed in this Q&A article: What antibody panel is available to use with the assay?
